we extracted vacuoles sap of guard cell and epidermal cell under a microscope and observed the distribution of the particles in vacuoles sap from photos taken under a electron microscope, we founded great difference in shape, dimention and density of particles in guard cell vacuoles and epidermal cell vacuoles. And what's more, these two kinds of particles response quite differently to pH control: there was almost no effect of pH 5.3 and pH 5.8 to the dimension and distribution density of the particles from epidermal cell vacuoles; however, pH 5.8 induced obvious increases in the dimension and distinct decreases in the distribution density of the particles from guard cell vacuoles, and no obvious changes had taken place to the particles from guard cell vacuoles after pH control of 5.3. The non-specific proteinase (Proteinase K & Pronase) cuold make a disapperance of the particles from both guard cell vacuoles and epidermal cell vacuoles. So the particles in vacuoles of guard cell and epidermal cell proved to be protein. We used the ratio method of dual fluorescent emission of chemical pH probe BCECF with single excitement under laser confocal microscopy and founded that there was a basification about 0.5 (5.2→5.7) units of pH in guard cell vacuoles and a acidification of about 0.3 (5.6 → 5.3) units of pH in vacuoles of epidermal cell which is close to a stomata during stomatal closure induced by ABA.This result may due to H~4 had moved from guard cell to epidermal cell.
