Thymosin α 1 (T α 1), a 28-amino acid peptide with an isoelectric point of 4.2 and a molecular weight of 3108 Da, is a peptide hormone secreted by the animal or human thymus organizations. T α 1 is a biological response modificator, mainly as the enhancer of T lymphatic immune system for the patients with immune deficiencies or constrained immune system. When patients were infected with certain viruses such as HIV, HBV or treated with radiotherapy or chemotherapy, the immune system tends to be compromised and even destroyed and can not kill cells contain a virus or cancer cells. T α 1 can restore the function of T lymphocyte , induce the proliferation of the mature T cells, promote the lymphocyte to gather around cancer and pathogeny tissue and accelerate the generation of lymph factors and lymphatic factor receptors, thus greatly enhance patients' resistance to the virus and cancer.Because of its length(28aa),the T α 1 is difficult to synthesize by genetic engineering methods;the current chemical products of T α 1 are expensive;In addition, we can extract T α 1 from animal thymus, but the purity and activity are lower so its clinical application is limited.To explore the possibilities of mass production of T α 1 or similar biological reactive product, we designed and constructed the fused expression plasmid of pET28-a-T α 1 and transfected it into E.coli BL21, then culture the engineering bacteria and purefy the fused protein, at last we measure the activity of fused protein.
