Malaria is a kind of terrible disease which can cause death of thousands of people, Malaria is caused by the Plasmodium,The African malaria mosquito(Anopheles gambiae) Plasmodium falciparum is a highly anthropophilic malaria-transmitting vector, Olfaction plays a major role in host finding and selection of malaria mosquitoes.Insect can perceive certain chemical volatiles in the environment ,as the chemical information sources for its behaviroral reactions ,such as to look for mate, food and reproduction place.odorant binding proteins (OBPs) are thought to deliver odors to olfactory receptors, and thus may be the first biochemical step in odor reception capable of some level of odor discrimination. And it's important for insect to transfer odor information in the environment and give rise to certain behavior response.in this paper,the expressing vector of Odorant Binding ProteinAgOBP48 in the Antenna of Anopheles gambiae was construted ,and AgOBP48 gene was expressed in Escherichia coil.The PET/20b-AgOBP48 was given by Dr Jing-jiang Zhou in Rothamsted Research United Kingdom, The aimed fragment was obtained by two restriction enzyme Ndel/EcoRI form PET/20B-AgOBP48 and further cloned into PET-28a(+) vector respectively.The aimed fragment was digested by two restriction enzyme Ndel/EcoRI and then was joined with pET-30a (+) plasmid which was digested by Ndel/EcoRI too.And then constructed into expression vector pET30a (+) for overexpression in prokaryotic cells. Structural analysis showed that the full length of matureAgOBP48 open reading frame (ORF) is 536bp, 172 amino acid residues were encoded.
