Selecting 13 high/low-risk genotypes HPV as targets and simulating clinical situation of single infection and multiple infections, a new method is established for HPV genotyping based on multiple suspension array. The samples of condyloma acuminatum samples and cervical erosion are genotyped using multiple suspension array.HPV plasmids recognized internationally as positive standard, HPV genotyping methods is constructed. The conditions is including common primers designing, probes designing and detection. PCR system and hybridization have been optimized, The results of HPV plasmids prove that the method can genotype correctly. In order to establish diagnostic standard, the discriminant function was set up using discriminant analysis of medical statistics. Discriminant function is tested with data of standard HPV plasmids detection and the correct rate is more than 96.7%. Compared with the commercialized kit using reverse hybridization, the consistent rate is 82%. The condyloma acuminatum samples and 100 cervical erosion samples from hospital was genotyped based on multiple suspension array, 79% positive condyloma acuminatum samples and 9% positive cervical erosion samples were detected.Compared with reverse hybridization in HPV genotyping, multiple suspension array is easier in operation with high throughout, sensitivity for genotyping and have more advantage and development space in clinical application. The genotyping results of clinical samples using multiple suspension array have shown great importance in disease forecast and epidemiology research.
