Thymosin α1 (T α1) is a biological response modifier with plenty of biological functions, such as antiviral action, and it can kill tumor cell and enhance immunity and so on. It is of great value to the clinical application and has been applied on the therapy of chronic hepatitis and cancer and so on. There are three preparation methods of it: distilling from animal tissues, synthesize and gene recombination. The amino sequence of synthesized and crude T α1 is identical, but the production cost of synthesized T α1 is much higher and the process will bring great pollution to the environment. Now the price of import synthesized T α1 is as high as 900RMB/1.6mg, so a lot of patients can not afford the high medical fee. T α1 is very difficult to be expressed directly because the polypeptide of it is very short. So it is often been expressed together with other proteins, such as ribosomal protein and so on. Then T α1 is isolated from the fusion protein by CNBr or hydroxylamine. But the chemical medicaments are often virulent. So the production will prick up the pollution of the environment and bring damage to operators. Gene recombination T α1 is in experimentation now. The crude T α1 have advantages over the two others. The price is very low and the production process is safe to the environment. Most of all, it has crude activation and is secure to the patients. But the problem is the content of T α1 is very low(often below 0.61%). In order to obtain high percent of T α1, we make this study. The research includes two schemes. The first one is, we purchase the crude T α1 and the HPLC analysis shows the content of T α1 is about 0.31%. Then it is purified by anion exchange chromatography and the content has increased to about 7%. The second one is, we purchase new born calf thymus and distill T α1 by low PH preparation method. The purity of T α1 reaches as high as 6.7% by HPLC. Then it is purified by isoelectric precipitation and anion exchange chromatography respectively, and the purity is 12.96% and 9.61%. The result is 14.46% by the method of isoelectric precipitation integrate with anion exchange chromatography.
