Study On Purifucation And Properties Of Superoxide Dismutase From Erdos Plateau Alkaline Lake Spirulina Platensis

Superoxide dismutases are a family of metallic enzymes essential for the survival of aerobic cells; they scavenge the superoxide radical, which appears to be a toxic species of oxygen. SOD catalyzes the dismutation of the superoxide radicals to hydrogen peroxide and oxygen. Now, superoxide dismutase has been isolated from germs, algae, high plants, animals etc. and applied to the daily life, such as the cosmetics, medicine, food, morbid examination of persons, animals and plants.Four strains of Spirulina were found in the alkaline lake of Erdos plateau in 1996 by Qiao chen, the professor of Inner Mongolia Agriculture University. One of these strains was Spirulina platensis, which was applied to the industries and was studied to find out that it would be an important spirulina strain resource for the developing of spirulina industry in North China as its low-temperature-resistance and thermo-broadness.In this paper, S₁ SOD was extracted and purified using Spirulina S₁ as materials and it's characteristics were also studied and contrasted with the SOD of Spirulina S₂ from the Chad lake of Africa and that of Spirulina S₃ from Sosa lake of Mexico, not only to reveal the SOD differences between these Spirulina strains, but also to lay the foundation for the application and developing of the Spirulina from the location. Extraction and purification of SODThe purified SOD was prepared using Spirulina S₁ , S₂ and S₃ as materials respectively in four-step processing in turns, ultrasonic breaking, fractionally (NH₄)₂SO₄ salting, DEAE-FF ion-exchanging and Sephadex G-100 gel filtrating.
