Objective: This research aims to propagate and save these two gene knockout mice as well as give them genetype authentication and study their biological properties. Methods:We identify the genetype of the mice through PCR, breed and save the heterozygous mice. Meanwhile we observe and examine the reproduction, growth curvature, physiological and biochemical index and viscera changes of different genetypes. Results: There are no significant differences in the reproduction, growth curvature as well as viscera structure and weight of different genetypes . There are no obvious difference in physiological and biochemical index except CK in heterozygous of the1a-hydroxylase gene knockout mice compared with that of the wild type. In VDR-KO mice ,white cell count of homozygotes increase significantly, mainly neutrophil granulocyte .and some biochemical indexes also increase such as ALP,ALT,TG,CHOL,LDH. Conclusions: PCR can be used to identify genetype of the geneknockout mice. In the1a-hydroxylase gene knockout mice and the VDR-ko mice, there are no significant differences between the wild mice and the heterozygous ones. However homozygotes of VDR-KO represent the characteristic of vitamin D-dependent rickets.
