The coculture of different generated endometrial cells of bovine during early to mid-luteal phase at Ya'an local abattoir (including the primary culture endometrial cells group,the early passage(3 generation) endometrial cells group, the late passage(7 generation) endometrial cells group and the early post-cryo-thawed endometrial cells group monolayer) and mouse embryos(collected after superovulation, mouse developed by Animal House of Huaxi) was detected by graded embryos on the basis of morphology, then the medium culture alone was the control group. Based on the detection of the morphology of embryos, the effects of different passage endometrial cells on overcoming the developmental block and embryonic development of mouse in vitro were analyzed, so we could deduce the mechanism of coculture, to prepare for the study of bovine autologous endometrial cells and embryos in vitro fertilization coculture, furtherly to increase the pregnancy rate that with in vitro fertilization-embryo transfer for bovine embryos development on autologous endometrial monolayer cells.1. After coculture and medium culture alone for 24h, some of the mouse embryos stopped in two-cell stage, but the number of embryos that sttoped in two-cell stage in the coculture groups were remarkably less than that in the control group (p < 0.05), No differences were found among the coculture groups. The blocked embryos did not cleave after 24h. In conclusion, the bovine endometrial cells could improve the developmental block in 2-cell stage efficiently.2.
