DNA is the main genetic material of the organism. Protein has important physiological function. The research about DNA and protein can reveal the magic of the organism. detection for the structure of the nucleic acid and protein, and the research about the interaction of the anticancer drug with nucleic acid. For these aims, the fluorescent probes play a important role.So, we focus on the fluorescent probes that can be useful for the determination of the DNA and protein in the biological samples. We hope to find some excellent fluorescent probes to determine DNA and protein. we can do more works about DNA and protein.Three aspect research have been carried out in this paper: Part 1: A novel fluorescence agent 2– methyl– 12 H– Benzo [b] [1,10] phenanthro– lin– 7 -one (m-BPO) was synthesized, and its molecular structure had been confirmed by IR, UV,. The fluorescence characteristics of m-BPO were investigated in detail. It was found that DNA had the ability to quench the fluorescence of m-BPO at pH7.4 Tris-HCL buffer, and the quenched intensity of fluorescence was proportional to the concentration of ctDNA. Based on this fact, m-BPO had been used as the fluorescence probe for detection of ctDNA. Under optimal conditions, the calibration graphs are linear up from 1.5 to 15.0μg/ml for ctDNA. The corresponding detection limit is 0.05μg/ml for ctDNA. Interference of different foreign substances was investigated at the same time. The interaction mechanism for the binding of m-BPO to ctDNA was also studied Part 2: It was found that BSA had the ability to quench the fluorescence of m-BPO, under pH7.4 Tris-HCL buffer , the quenched intensity of fluorescence was proportional to the concentration of BSA. Based on this fact, m-BPO had been used as the fluorescence probe for detection of BSA. Under optimal conditions, the calibration graphs are linear up from 0.05mg/ml to 3.0 mg/ml for BSA..
