Embryonic stem cells have the totipotency of differentiation. Investigate and use the ES cells is one of the hotspot of biology engineering. Most of the ES cell lines are developed from129, C57BL/6J, BALA/C strain. Researches of establishing the ES cell line of Kunming Bai/strain mouse were rare. As a widely used laboratory animal, investigate the methods of building its ES cell line has extensive application.This study mainly investigated the method of ICM isolation, the cell line passage to the next generation, and the culture effect of ES cell clones in the RH-CM. Different feeder kinds' impact on ES clones were also compared. The results showed that the multi-clonal antibody of rabbit-anti-mouse can effectively destroy the trophoblast cells of mouse blastocysts. The passage of inner cell masses isolated by immuno-surgery was best when cultured onto PMEF feeder & LIF system. The ES clone have passed to the 8th passages in vitro. The ES cell has the ability to form nest-like clone, the boundaries of ES clones are clear, the ES cells clones are very compact. They were AKP positive in colour of deep brown, and still have pluripotency to differentiate into many cell types. After refrigeration and deposit at liquid nitrogen, the ES cells were thawed and survived in vitro.This study also gives a summary include the process of ES cells development, progress on ES cells technology, existing problem and the prospect of its future.
