This study was carried out with two parts of experiments.In experiment 1 , the purpose of the study is to investigate the characteristic of microsaterlite and their application in Macaca such as Macaca Fascicularis and Rhesus so as to provide a scientific basis for the conservation and utilization of Macaca genetics.By using the on-line bioinformatics software of the repeatmasker on Internet, the ESTs database of macaca was screened. Five primer pairs were designed from microsatellites using computerprogram Primer Premier (Version 5.0) and Oligo 5.0. In these five microsatellite loci, the alleles numbers distributed from 5 to 11, and the range of these alleles were 150bp to 320bp and similar with their predicted size. The result of the whole experiment showed that 4~ 5 of these 5 microsatellite loci were suitable for genetic analysis.EB staining method, through this method was available on separation of allele of microsatellite. The agarose gel electrophoresis run in 2.0~3.5% agarose gel for 2.5~5 hours by 65~100 Volt. The separate allele can be reclaimed and purified separately ,and then sequenced.The result showed that some microsatellite DNA composes a composite complex. The pilot study of identifying the relationship between parent and offspring by using microsatellite marker technology showed that it can be glancingly identified from agarose gel.
