Transgenic animal is a new born bio-technique, which is the development and extension of general molecule biology and gene engineering. With the appearing of transgenic animal, it is possible that people can rebuild the genome of animals by gene recombination and contrive research on relative gene's structure and function in viviperception. This is a multidimension system of gene research with regard to providing new approaches and thoughts from molecule to individual arrangement. Transgenic animal reflects the complexity of nature, opening up avenues of life research previously impenetrable, and having a splendid application as well.C-reactive protein and serum amyloid p component are members of the pentraxin family of oligomeric serum proteins which have been conserverd through evolution. Both have pentameric structures and both play complex roles in the innate immune response, especially serum amyloid p component is an important protein in tissue damage, inflammation and amyliodosis.Human genes have much higher with mouse allelomorph,meanwhile, human diseases have more competitive with mouse disease models, which give a new research method from human diseases to target genes. The approach of gene micro-injection was firstly used by Clark's research team in Britain, aiming to improve the expression level of foreign gene by its cooperation action. Now, in the paper, mouse SAP gene was micro-injected into mice in order to study the gene function.Firstly, according to the sequence of mouse serum amyloid p component gene in GeneBank, two primers were designed and amplified mouse SAP gene by PCR. Secondly, PCR products were subcloned into pCMV-Tag5A vector and transformed E.coli.DH5αhost strains. Correct clones were selected and plasimd DNA was extracted and digested with BamHI and XhoI.
