Musk deer (Moschus spp.) is the most important resource animal of China, and the first class of "Key"species of wildlife protected by Chinese. Due to poaching for musk pods and the disturbance of habitat, musk deer populations are redued sharply, and are becoming seriously endangerous. Now there are many researches in taxology, histology, physiology of musk deer, However, little research about molecular genetics of musk deer. Our libertary do some researches in genetics of musk deer .Microsatellite DNA is co-dominate genetic marker, it can fealty reflect personal feature, the parentage relationship, it have been wildly used in human and livestock. In this time, more microsatellites loci in musk deer were found and the characterizes of these loci also analyzed, The genetic diversity of the two captive musk deer populations were analyzed also.In this research we used the method of Bloor et al.(2001) and make some changes. We chose two oligonucleotide (CAA)_n and (AGC)_n which have higher frequency than others in mammal. [Ga'bor To et a.2000] According to the strong affinity between streptavidin and biotin, this two oligonucleotide labeled with biotin, respectively. This two probes were then reacted with streptavidin-coated magnetic beads. The genomic DNA of forest musk deer were digested with restriction enzyme Sau3AI. The 400-900bp DNA fragments were isolated, ligated with linkers Oligo A and Oligo B, then annealed with streptavidin-biotin-probe...
