In this paper, the immobilization of the DNA probe on the solid electrode was investigated mainly using electrochemical method. With methylene bule (MB) as hybridization indicator, the electrochemical DNA biosensor was prepared and used to detect the products of the transgenic plant.The paper is made up of four parts. Firstly, a summary referring to the development of the transgenic plant, and the methods of the immobilization of the DNA probe on the electrode and the hybridization indicator was reviewed. The second part and the third part are the investigation on the role of aluminum ion and zirconium ion during the course of the immobilization of the DNA probe. After the understanding on the changes of electrochemical parameters of MB such as the redox peak currents, potentials, at different modified electrodes, we found both of them improve the properties of the electrode significantly, and make the immobilization and hybridization easily on the surface of the electrode. The conditions for the immobilization and hybridization of the DNA probe were optimized and the biosensors were used to detect some transgenes in the transgenic plants.
