As one of the most significant methods for understanding the biological function of protein, the interaction of proteins is extremely important in the research of functional genomics. The interaction exists within the vital movements of each cell in the body. Many phenomena in biology, such as apoptosis,cell cycle control,signal transduction and canceration and etc, are all regulated by the interaction of proteins. There are various ways to investigate on the interaction of proteins and those commonly used are yeast two-hybrid system,affinity chromatograph,immunoprecipitation,protein cross-linking and GST pull-down and etc, among which yeast two-hybrid system is the most rapidly developed and widely used method.Tec is a kind of non-receptor tyrosine kinase which was firstly isolated from the liver cancer tissues. It participates in signal transduction of many cytokines,and it is an important molecule in the signal transduction of many cytokines, assistant factors and the receptor PTK. Tec participates in signal transduction in response to virtually all types of extracellular stimuli which are transmitted by growth factor receptors,cytokine receptors,G-protein coupled receptors,antigen-receptors and integrins and etc. In the meantime, Tec can be regulated by many non-receptor tyrosine kinases such as Src, Jak, Syk and FAK family kinases. Furthermore, Tec is concerned with many of the major signaling pathways including those of PI3K, PLC and PKC. Research shows that Tec is the key joint molecule of protein kinase in the signal transduction pathway of activation,growth,proliferation and maturity of hematopoietic cell and immunologic cell. Since kinase domain(KD) of Tec is one of its featuring structures, we can have a deep understanding on the functions of Tec and thus it can provide important clues to clarify the position and effect of Tec during the process of signal transduction pathway of proliferation,induction and differentiation through research on this domain and its interacting protein molecule.We use Tec kinase domain as the bait protein in our preliminary work and take advantage of the technique of yeast two-hybrid system to filter the cDNA library that was built from...
