| Objective By electrical stimulate adult rabbits' ventrolateral pontine to observe the effect on the respiration with phrenic discharge as the target. By microinjecting N-methyl-D-aspartate (NMDA) receptor agonist L-glutamate and antagonist 2-amino-5-phosphonovaleric acid (AP-5),α2 adrenergic receptor norepinephrine(NE) into the ventrolateral pontine,to observe the effects of the three kinds of drugs on the post-hypoxic frequency decline in later acute hypoxia, and then to investigate role of NMDA and a2 adrenergic receptors in post-hypoxic frequency decline.Meanwhile,we employed several immunohistochemical methods in our experiments in order to explore the function of tyrosine kinase pathway in the intracellular signaling transmission after NMDA receptors on ventrolateral pons have been activated by hypoxia. Methods 1 .Electrophysiological experiment:(1)Electrical stimulation: The experiment was exerted on 56 healthy adult rabbits ,parameters:frequency 100Hz,current between 20μA and 50μA,between 50μA and 80 μ A,width 0.3ms,trains 3s,after stimulation,L-glutamate microinjected to observe the effect on respiration with phrenic discharge as targets.(2)Microinjection:The experiment was conducted on 45 healthy adult rabbits and 30 results were available.Experimental rabbits were divided into five groups as follows, and rabbits in different experimental groups received microinjection of different... |
