Aim : Glucomannan is quantified by the method of phenol-sulfuric acid; Aminoglucomannan is produced with ultrasonic cavitation that spur the hydroxyl group of glucomannan on to be ammonified in the ammonia. The proportion of aminosugar in the aminoglucomannan is determined; The optimum conditions is explored under which aminoglucomannan is marked with fluram, the fluorescence properities of the product(Fluram-AGM)is also explored; Fluram-AGM transported into cell across cell membrane is traced for the study of the transport of aminoglucomannan across cell membrane. Finally, it is confirmed that the aminoglucomannan can be transported into cell across cell membrane.Method : 1. Quantification of glucomannan: Firstly, the standard glucose is dissolved in the distilled water as a series of standard solution. The glucose in the standard solution reacts fully with the phenol and sulfuric acid which are added into standard solution, and the absorbance of solution is measured with spectrophotometer on condition that the wavelength of incident ray is 490nm.
