The abnormality or absence of human tooth is prevalent now. The final purpose of tooth regeneration by chimeric tissue recombination is to research the odontogenic potential or odontogenic competence of stem cells from human's dental pulp,and preliminarily set up the technology model of human tooth regeneration in vitro.This will establish theoretical and techynological foundation for the further research of regenerating tooth germ or tooth which can be transplanted in vitro.This study utilized the culture of cell in vitro to isolate and culture SHED and DPSCs.And we devoted to researching if the stem cells from human's dental pulp had odontogenic potential or odontogenic competence by tissue recombination,organ culture and tissue morphological analysis.The content and results were as follows: First of all,we made chimera which recombinated SHED or DPSCs with E10.5 non-dental epithelia of mice,and we found that SHED or DPSCs can't induce non-dental epithelia to form tooth.which indicated that SHED or DPSCs didn't have the odontogenic potential; secondly,we made chimera which recombinated SHED or DPSCs with E10.5 dental epithelia of mice,and examined the morphological change of different developmental period of the chimera.The result was that it rarely can form tooth or develop into tooth germ,it illuminated that SHED or DPSCs didn't have the odontogenic competence directly in vitro;thirdly,we mixed SHED or DPSCs with mice E13.5(mE13.5) molar mesenchymal cells depending on different proportion,then recombinated it with mE13.5 dental epithelia,and we found that SHED or DPSCs can be induced to form tooth, which illuminated that SHED or DPSCs had the odontogenic competence indirectly in vitro;the last,we recombinated SHED or DPSCs with E10.5 dental epithelia of mice and researched the regeneration of tooth by the assistant induction of foreign growth factors such as BMP-4 or FGF-8, finally we found it can form tooth,which indicated that SHED or DPSCs can be induced to form tooth by dental epithelia of mice,under the assistant induction of foreign growth factors.Our result ultimately set up the technology of human tooth regeneration preliminarily by stem cells from human's dental pulp in vitro.
