Study On The Transcriptional Regulation Of KRAB Zinc Finger Protein Gene ZNF331 | Posted on:2007-09-03 | Degree:Master | Type:Thesis | Country:China | Candidate:Y J Chen | Full Text:PDF | GTID:2120360185993975 | Subject:Medical Genetics | Abstract/Summary: | PDF Full Text Request | Part I ZNF463 gene is a spermagenesis related gene which wasisolated and characterized by our laboratory recently. It is one of the spliced form of ZNF331 gene. To study the localization of the ZNF463 protein, green fluorescent protein(GFP) was used as a marker. The full length cDNA of human ZNF463 obtained by RT-PCR from normal human testis tissue was inserted into mammalian expression plasmid pEGFP-Cl. The construct was then transfected into Cos-7 cells by liposome. Results showed that the fluorescence located mainly in the nuclei of the cells when transfected with constructed recombinant pEGFP-ZNVF463. While after transfected with vector pEGFP-Cl, the GFP expressed and located over the whole cell . Thus it could be concluded that the transfected Cos-7 cell line can express human ZNF463 protein, and this protein localizes mainly in the nucleus, which is consistent with its being a member of a family of transcriptional factor.Part II ZNF331 gene is a newly isolated thyroid adenoma related gene. Its abnormal expression might be one of the molecular...
