Myostatin, a member of the transforming growth factor-beta (TGF-beta) superfamily, is a potent negative regulator of skeletal muscle growth,and myoblast could be proliferation and hypertrophia when myostatin lossed activity. Recently ten years,study have got great progess on structure of myostatin, disposition chromosomal and protein expressed,but it's not agree in records about in Distribution organism .However, myostatin's mechanism of action and regulation have not been understand.For the reason , The objective of this study was to produce a monoclonal anti-myostatin antibody and to examine the tissue distribution in porcine.Prokaryotic expression and purification or porcine myostatin C-terminal polypeptide. Recombinant engineering bacteria MSTN-PGEX-4T-1 had been induced by IPTG,and all thallus is broke into pieces by hyperacoustic, then GST affinity chromatograph purified GST-MSTN ,and C-terminal MSTN purified from GST-MSTN by thrombine . 12% SDS-PAGE and 15% SDS-PAGE analysis of the purified GST-MSTN and C-MSTN protein preparation revealed the presence of two protein species of 38 kD and 12.5 kDa.It's suggested that these purified protein conformed with theory .Purified GST-MSTN fusion protein as a immunizing antigen to...
