Object: This study researched the effect of anger on cytokines and related enzymes in brain tissue of mouse to explore the acting mechanism of sub-health caused by anger. We detected the contents of IL-1B and TNF-a in brain tissue of angered mice with enzyme-linked immuno-sorbent assay (ELISA). We also detected the activity of Superoxide Dismutase (SOD), Acetylcholinesterase (AchE) in the tissue. Furthermore, we treated the mice with Xiaoyaosan, an extract of seven traditional Chinese medicine herbs, Bupleurum chinense DC, Paeonia lactifora Pall , Angelica sinensis (Oliv.) Diels . Atractylodes macrocephala Koidz, Poria cocos (Schw.) Wolf. Zingiber officinale Rosc. Glycyrrhiza uralensis Fisch, and detected the cytokines and enzymes. This would help us to analyze the therapeutic efficacy and mechanism. It can be foundation for the prevention and treatment.Method: We divided the mice into health group and model group. The mice of model group were enraged with nipping their tails. After intelligence test, mice of the two groups were killed. Their brains were taken out, and homogenized. The homogenates were centrifugalized under 4℃,3500r/min. IL—1βand TNF—a in the supernatants were quantitated with ELISA. We also detected the activity of SOD and AchE of the supernatants. Mice were divided into 3 groups respectively by different dose and treatment time when we treated them with Xiaoyaosan. The same targets were measured.Results: Learning and memorial ability of model group declined. The content of IL-1βand TNF-a increased. The activities of SOD and AchE didn't have any obvious change. After treatment with Xiaoyaosan, content of IL-1βand TNF-a reduced, and the learning ability improved.Conclusion: The changes of nervous system function induced by anger may have relation to chronic activation of immune and HPA axis dysfunction. They have little relation to activity of SOD and AchE. Xiaoyaosan could improve the state of sub-health caused by anger.
