In this thesis, an oligonucleotide DNA microarray was designed for Helicoverpaarmigera single nucleocapsid nucleopolyhedrovirus (HearNPV), and used toinvestigate the global viral gene expression of HearNPV.Chapter 1 was a literature review. It contained the introduction of baculovirus, areview of microarray including the gene noise and signal processing used inmicroarray. And it introduced the purpose of this thesis.In Chapter 2, a oligonucleotide DNA microarray containing all the potential (135)viral genes of HearNPV was designed and validated.In Chapter 3, total HzAM1 cellular mRNAs extracted between 1 and 144 h postHearNPV infection were detected by the microarray. Gene was divided to 3 classaccording to their expression time. The results revealed that there were two periodsduring the transcription, the first period was depended on the host RNA polymeraseand the second period was depended on the viral RNA polymerase. The expression ofthe structural proteins appeared to be ordered into BV (Budded Virus) related or ODV(Occlusion Derived Virus) related. To gain further insight into the regulatory network,a SVD method was used to screen the important factors in transcription and ie-1 andcg30 was found to be most important regulation gene.In Chapter 4, ICA and wavelet methods were used for data mining to reveal thelarge-scale genome location-dependent expression patterns of HearNPV. Aperiodicity of about 25-kb were detected and the homologous regions (hrs) of the HearNPV were found to be located at the symmetry centers of the periodicity. Itindicated that there were 5 DNA-reactors in HearNPV genome based on hrs. GC-walking analysis of the genome DNA revealed the correlation between theDNA-reactor fractal dimensions and the transcption patterns.
