Abies yuanbaoshanensis (Pinaceae) is a highly endangered fir species and only distributing on Yuanbaoshan mountain, Guangxi, China. It is listed in the China Plant Red Data Book as class I, and is an IUCN protected key species. Its stenotopic distribution center is an area of about 20 ha, at 1900~2000m on the Lanping Peak of Yuanbaoshan Mountain, with less than 1000 individual plants experiencing very severe conditions. Abies yuanbaoshanensis community, therefore, is clearly a key biodiversity group in China.In this study, 125 individual's young leaves of mature trees were selected from three districts in population and cpSSR and SSR were adopted as the molecular markers to infer the genetic diversity and genetic structure of Abies yuanbaoshanensis. An understanding of genetic diversity and population genetic structure of Abies yuanbaoshanensis provided a scientific basis for worked out scientific plans and programs for the protection and adopted a sequence of measures for effective protection of the rare and endangered species from conservation Biology .The conclusions of study on Abies yuanbaoshanensis by two molecular markers thereinafter:It was revealed that total number of haplotypes was 14 in the population of Abies yuanbaoshanensis by cpSSR marker. The effective number of haplotypes was 6.9168 for the population and the result of heterozygosity based on frequency of the 14 haplotypes was 0.8554. It also revealed that four haplotypes of the 14 haplotypes were common existed in population of Abies yuanbaoshanensis .Using the information from 7 microsatellite to estimate level of genetic structure of Abies yuanbaoshanensis .The results based on SSR marker revealed that the observed alleles and effective alleles were 16 and 9.7073, the observed heterozygosity and expected heterozygosity were 0.2834 and 0.3088, the heterozygote deficit within population (Fis) amounted to -0.041. The results of the expected heterozygosity and the heterozygote deficit within population (Fis) indicate that Abies yuanbaoshanensis has a low level of genetic diversity but a trivial excessive of homoplasy.The results of NJ analysis based on Nei&Li genetic distance and principal components analysis (PCA) based on SSR marker for population of Abies yuanbaoshanensis were revealed that a high degree of genetic similarity but low level genetic diversity were existed in the population.In summation, a result of low level genetic diversity and high degree genetic similarity existed in the population was revealed by study on genetic diversity and genetic structure of Abies yuanbaoshanensis which based on cpSSR and SSR marker. The finding provides valuable information for formulating future conservation programs for the species. It is suggested that all individuals of natural population should be protected and the distribution region of Abies yuanbaoshanensis should be given sufficient attention and effort of protection.
