The puerarin is a kind of effective component which is abstracted from puerarain lobata or puerarin thomsoni,they are leguminous plants,or abstracted from the roots of the sibling plant.Puerarin is one of the maximum chemical compound ingredients in the osajin which is abstracted from the puerariae root.The predecessors have studied a lot to the pharmaco-analysis of the puerarin from clinical.They found that the puerarin is a effective medicine for the endeictic of high blood pressure,hyperlipemia,coronary artery disease,angina cordis,cerebral thrombosis,and at the same time,the therapeutic efficacy of puerarin is very effective to preserve the arterial blood vessel endothelial,to treat the disease of artherosclerosis,to prevent the thrombogenesis.It is the first time to use the microelectrode technique to study the puerarin,s pharmaco-investigation,and especially the tosicoadverse reaction mechanism of the puerarin has not been studied by the predecessors.So,we study the puerarin,s pharmacologic action from the cell electrophysiological level.We hope to offer some theory evidence for the use of the puerarin,s clinical application.The main method we use in our experiment is microelectrode technique.The steps are as the following:1,Preparing the cerebrospinal fluid one hour before the experiment,take some of them for cryopreservation;2,Without paralyse,cuting down the rat head quickly and dislodge the hippocamp part which is used in the experment in freezing liquid,and trim the hippocamp distrct.3,Fixing the hippocamp tissue on the vibratome,to cut down a brain slice with 300μm,use the haustorial tube to move the slice to the preincubate dish,cultivanting it for one hour.Then move it to the record incubation chamfer.4,Preparing the glass micloelectrode and fill it with NaCl,the corcentation is 3mol/L.5,adjusting the recorder,after cultivate the brain slice in the incubation chamfer for 2 hours,the expermentize can be gone on.6,In order to ensure the accurate of the experimental result,use only one medicine concentration for one brain slice in every experiment.The main content of the study are those as following:1,The status quo of the puerarin,s pharmaco-study.In this part,the main contents refer to the resource overview of the puerarin,the advancement of foundational study and clinical research of the puerarin, the overview of pharmaco-function and the adverse reaction caused by the puerarin,and so on.After the work of this part,from the summary we can know that the active compound of puerarin can promote the hemokinesis of the heart and the amphiblestrodes,relax the spasmolysis of smooth muscle,anti-promotion cancer and induce the cancer cell differention,drop the blood glucose and blood pressure,anti-oxidation,relieve fever,alleviate a hangover and increase the faculty of memory,as well as the function of the puerarin can strengthen the body,s immunity and so on.2,The spontaneous discharage types of the hippocampal nerve cells in the rat limbic system and the study for the neurotransmitter,s effect to the nerve physiological function.According to our study,we found that the discharage regulation in rat,s four districts(CA1,CA2,CA3,4CA) of hippocampal exist some kinds of difference.There are six main discharage regulation release types in the hippocampal neuronal .They are periodicity volley release type,paroxysm and random release type,periodicity volley and sing admixture release type,single regulation release type,slow speed release, release close-up quickly type.The purpose of our study is to find the neuron,s basic spontaneous discharage regularity and the difference of the electrophysiology of the different part in the hippocampal.The experimental result shows that the amplitude and frequency of CA1 discharage release are much more ideal than the ones in the other three parts.The ones in CA1 are easy to be controlled and analyzed,the ones in the other three parts are fluctuate now and then,it may create some kinds of interference to the analyze of the experiment result.We perfuse the routine tool medicine as Glumatic acid(Glu),Acetylcholine(Ach), γ-aminobutyric acid(GABA), 5-hydroxytryptae(5-HT) and Glycine(Gly) to the brain slice each one which are the ones that spontaneous discharge release much more steady,the result we find that the Glu,Ach and 5-HT have some evident periaqueductal gray stimulation,the GABA and Gly can depress the hippocampal neuronal,s discharage significantly.The effect mechanism may connect with the corresponding receptor which exist in the hippocampal.3,The effect of the puerarin to the electrophysiology of organization CA1 of the rat,s limbic system.This part is the main body of the experiment,in this part,the medicine we use are puerarin,Ach and L-Glu. First,we perfuse puerarin to the brain slice which electricity activity is better,one concentration per one brain slice,the result shows that the puerarin has conspicuous depressant effect to the hippocampal,s spontaneous discharge act,the mian abaissement modality is to cut down the frequency,but the effect to the amplitude is very little.Second we use excitability medicine to provocat the nerve,s causality discharage,after 1 to 2 minutes,perfuse the brain slice with a certain concentration of puerarin to be abaissement(to interrupt),in result,we find that some kinds concentration(such as 1:1000) of puerarin have prominen depressant effect to the provocated discharage of the hippocampal nerves. The depressant effect is very quickly,the frequency can be shut down sharply.The pharmaco- function effect of puerarin has a minimum concentration clicker value,but the maximum clicket value(lethal concentration) is not evident.The concentration dependent to the hippocampol is strong.According to our empirical study,we conjecture that there must be some homologus receptor exist in the hippocampal,and the main one isβreceptor.
