As it is indicated from the passing data, the most familiar human cancer is caused by p53 mutation, p53 gene is the most important member in all the anti-cancer genes, thus causes a lot of attraction.Expression vector PET-32a of p53 gene was constructed, induced the expression of P53 protein by IPTG in BL21. Then the protein was purified to effect on tumor cells.The methods of MTT, clone formation, FACS, DNA ladder etc. were used to detect the effect of P53 protein on K562 cells. And we also use the magnetic nanometer particulate to link with the protein (MNP-NH2-P53). Then we use the AFM to help us locating the p53 protein in tumor cells.From all the methods we can come to a conclusion that this protein had the inhibitory effect of cellular proliferation and induced the apoptosis of tumor cells. This experiment provides a dependable data in looking for a new anti-tumor genetic engineering medicine.
