Objective: To bolt RAPD and Microsatellite DNA marker from Carassius auratus Red Variety Inbred Strain C1HD, and obtain their information of genetic diversity.METHODS: Using 20 random primers from 80 primers and 9 microsatellite primes, 8 samples of inbred strain C1HD and common Carassius auratus red variety were analyzed by random amplified polymorphic DNA (RAPD) and Microsatellite DNA, respectively.RESULTS: RAPD analysis showed that a total of 144 loci were amplified from Carassius auratus inbred strain C1HD, of which 31 loci(21.5%) were polymorphic. 162 loci were amplified from common population, of which 62 loci(38.3%) were polymorphic. The size of all loci ranged from 0.2kb to 3kb. A primer (S333) was observed to produce a special band, and this band could be used as a molecular marker for discriminating inbred strain C1HD red variety from common population. Based on genetic distances clearly a dendrogram was constructed using the UPGMA method. In the dendrogram, all individuals in inbred strain C1HD population were clustered into one group, and all the individuals originated from common Carassius auratus red variety population were joined into another group. The results by microsatellite DNA showed that the mean number of alleles of inbred strain C1HD population(1.444) per locus was lower than that of common Carassius auratus red variety population(2.111). The observed average observation heterozygosity value(H0) and expectation heterozygosity(He) were detected also lower than that of common Carassius auratus red variety population.CONCLUSION: There was high genetic similarity in the inbred strain C1HD Carassius auratus red variety. A band observed from primer(S333) by RAPD could be used as a molecular marker for discriminating inbred strain C1HD Carassius auratus red variety from common population.
