Commercial proteins produced by mammary gland bioreactor have been widely study inbiotechnology area. Research of mammary gland bioreactor began with transgenic mice,followed by preparation of large animal mammary gland bioreactors. Studies on mammary glandbioreactor of ruminants have a good prospect. Dairy cattle have high milk yield, high-expressiondairy cattle will have high yield and good benefit. However, dairy cattle are monotonous animals,have long production period and high cost. Some examples of transgenic goat mammary glandbioreactor succeed. Dairy goats have long lactation period and relatively low cost, it has greatvoluble in preparation of mammary gland bioreactors. In base of this can impulse thedevelopment of dairy cattle mammary gland bioreactor.Dairy goat mammary gland epithelial cells cultured in vitro can be greatly used in research ofmammary gland development, lactation mechanism and mammary gland bioreactor. Several dairygoat mammary gland epithelial cell lines do not have ability of lactating so can not be used as a cellpattern for research on lactation mechanism and mammary gland expression vector detection.Therefore, it is necessary for more study of dairy goat mammary gland epithelial cells cultured invitro and growth factors impacting on mammary gland epithelial cells to seek an effective methodfor dairy goat mammary gland bioreactor cell mode.We chose guan zhong dairy goat which have good lactation performance for this study.Mammary gland epithelial cells were obtained by digestion, cell were purified and verified. Adairy goat mammary gland epithelial cell line was established through cell culture system anddetection of cell biological characters. SDS-PAGE, biochemical determination of triglycerideand HPLC can be used for detection of milk protein, milk fat and lactose. It reflects lactationfunction of insulin and probating impacting on dairy goat mammary gland epithelial cells. This isvery useful for cell mode of dairy goat mammary gland bioreactor, and to provide experimentalevidences for dairy goat mammary gland development and lactation mechanism.Experimental results show: Purification dairy goat mammary gland epithelial cells wereobtained. Passage and resuscitation of cell can make the mammary gland epithelial cell lineconserved for a long time, a great quantity of mammary gland epithelial cells were obtained.Dairy goat mammary gland epithelial cells kept good growth state in culture fluid with additionof growth factors; cell could pass 30 generations and still remain good proliferation vital force.Insulin has no obvious impact on lactation function of mammary gland epithelial cells cultured invitro, while prolactin may induce secretion of milk proteins and lactose by mammary glandepithelial cells cultured in vitro.
