Fucoidans is a unique class of sulfated polysaccharide with many special activities.And oligosaccharides from fucoidans are more valuable than the whole fucoidans due to their low conglutination and high solubility.In this paper, samples are collected from water and in Zhengzhou in order to obtain bacteria producing fucoidanase.After enriching,separating,first-screening and second-screening,20 strains capable of producing high fucoidanase activities are separated and selected.Among them,the strain HNT01 has the highest activity and the fucoidanase activity is as high as 3.27U/mL after optimaling culture media.After the the colony features and the detection of 16srRNA are analyzed,the strain HNT01 is identified to be Pseudomona fluorescens.After single factor tests and orthogonal array,the optimal medium consisted of 2% kelp power,2%urea,0.1%K2HPO4,0.06%glucose.The initial pH of medium was 7.0. The volume was 100mL in 250mL flask.After shaken at 160rpm and cultured in this optimal medium for about 76 hours at 30℃,the fucoidanase activity is the highest.By studying characteristics of fucoidanase,a conclusion has been made that the fucoidanase has a high thermal stability and pH stability.And fucoidanase can retain itsactivity under 20-50 centigrade and within pH 5-7.At the same time,the study shows that The fucoidanase from HNT01 displayed an optimal temperature at 50℃and optimal pH at 6.0.
