H2S was considered as an endogenous gasotransmitter, which plays important roles in physiological and pathophysiological processes in cardiovascular systems. The mechanisms of its suppressive effect on heart function were poorly known. The aim of the present study was to observe the effect of H2S on heart function and investigate its mechanisms. METHODS: The effects of H2S on heart rate, coronary flow and heart function were observed in isolated perfused rat hearts. And the effects of H2S on ICa,L and IKATP were observed in isolated rat ventricular myocytes. RESULTS: 1. H2S can significantly suppress the heart function in the isolated perfused rat hearts; 2. H2S can significantly suppress ICa,L; 3. H2S can open ATP sensitive potassium channels. CONCLUSIONS: These results show that H2S can significantly suppress heart function. H2S may suppress the heart function by inhibiting ICa,L (decrease calcium influx directly) and opening KATP channels (decrease calcium influx indirectly).
