Study on molecule clone and expression of BMPs and them receptors, for make sure the reproductive function of BMPs and them receptors, and for clarify the mechanism of sterility and fetal from geng angle.In this study, we first cloned part of cDNA fragment of bovine BMP-15,GDF-9, BMPR-1B,BMP-4 gene by RT-PCR; then examined gene expression of BMP-15,GDF-9, BMPR-1B,BMP-4 gene mRNA at bovine different tissue by semi-quantitative RT-PCR. Methords: Firstly, search -ing for the BMP-15, GDF-9, BMPR-1B, BMP-4 gene sequencese of human, rat and sheep in Genbank, and designed the specific primers. Extracted total RNA from bovine ovary and amplified part of BMP-15, GDF-9, BMPR-1B, BMP-4 gene cDNA by RT-PCR, then reconbined these fragment into PMD18-T vector. After restriction endonuclease digestion and PCR analysis, the reconbined plasmids were sent to sequencing. The products of RT-PCR are the fragment of bovine BMP-15, GDF-9, BMPR-1B, BMP-4 cDNA. Secondly, According toβ-actin as inner Criteria, examined the possible expression of BMP-15, GDF-9, BMPR -1B, BMP-4 gene at bovine different tissue by semi quantitative RT-PCR (SQ RT-PCR).The result proved that BMP-15, GDF-9, BMPR-1B, BMP-4 gene mRNA all have expression in bovine ovaries, oviduct, uterus and kidney. The expression of BMP-15 gene in bovine ovaries and oviduct is highest; the expression of GDF-9 gene in bovine ovaries is highest, but lower in oviduct, uterus and kidney; the expression of BMPR-1B gene in bovine ovaries, uterus is higher than oviduct and kidney; the expression of BMP-4 gene in bovine ovaries is highest, and it is higher in oviduct than in oviduct and kidney.
