Coenzyme Q10, a kind of lipid soluble compound of quinone homolog, generally exists in the animal, vegetation and microorganism with microamount, it plays an important role as a member in the respiratory chain. Coenzyme Q10 exhibites excellent medical and physiological activities against various diseases, it is the natural antioxidant in vivo, activating agent of metabolism, it can improve the immunity of organism and have a general usage in clinic.At present, the Coenzyme Q10 yield was not universal very high, and the production of Coenzyme Q10 by microbe fermentation is the most hopeful method to be industrialized. In this paper, the Candida tropicalis APV.12 was selected as the fermentation strain, and the main research included that the effect of additives to the Coenzyme Q10 yield of Candida tropicalis and chosing and optimizing the extraction technology.1. This paper studied different breaking cell methods, the compound breaking cell was chosen; compared several commom extraction methods and the extraction by solvents was chosen eventually, and optimized extraction method. At last, the Coenzyme Q10 yield was improved from 13.3mg/L to 17.9 mg/L, adding 31.6%.2. The growth curve of Candida tropicalis has been studied, and the suitable fermentation conditions of Candida tropicalis were: temperature: 30℃, initial pH:6.0, inoculum size: 6%, and the best fermentation time was 50h.3. Tobacco, Carrot, Tomato, soybean oil, PHB, Phe, VB2, Met, Cys all can accelerate Candida tropicalis to synthesize Coenzyme Q10, especially Tobacco, Carrot and PHB, and these three additives compared to no additive , adding 43.5%, 31.4%, 29.3%.4. The compound accession of these three additives was studied by Statistics Analysis Software, and the result was that every one hundred milliliter fermentation culture medium added 0.085g PHB, 11mL Carrot, 0.21g Tobacco, the Coenzyme Q10 yield of three additive compound accession reached 24.9mg/L, and compared to no additive, adding 48.6%. However, the mutual effect of these three additives was obvious.
