Research On Fluorescent Multiplex PCR Of Canine's Eleven STR Loci

Short tandem repeat (STR) markers become the workhorse of many areas, such as genetics and forensic science. STRs are highly polymorphic DNA and capable of drawing gene map, guiding plant and animal genetic breeding and generating genotyping for parental identification, etc.In our study, we selected 11 highly polymorphic microsatellite loci PEZ1, PEZ2, PEZ3, PEZ5, PEZ6, PEZ8, PEZ12, FH2010, FH2054, FH2132 and FH2611, designed PCR primers, established a fluorescent multiplex system of 11 STR loci. Meanwhile, using this system , a total of 120 samples from police dogs was tested. It provided that the fluorescent multiplex system with 11 canine STR loci had been successfully constructed. The fluorescent multiplex system was of good productivity, good balance between different loci, no extra peak and no overlapping. And we obtained the forensic genetic parameters, the average heterozygosity (Ho) of the 11 STR loci in the dogs tested is 0.502, the average polymorphic information content (PIC) is 0.64, and the cumulative rate of exclusion is 0.9330621, and the cumulative rate of individual identification (DP) is 0.9999999. Hardy-Weinberg equilibrium test showed that PEZ3 and FH2010 in the German sheepdog deviated from Hardy-Weinberg equilibrium.The fluorescent multiplex system with 11 canine STR loci was successfully used in the ABI genetic analyzer detection platform to get the reliable data. The research of forensic genetics implies that it is an easily-regulated, low cost multiplex system with stable outcome, high sensitivity. Based on these advantages, it could be recommended to the practice of forensic genetics.
