Akebia trifoliate koiaz is a kind of high economic wild plants,and is being cultivated and planted as a new fruit resource.Its peels,whose weights account for 40-70%of the whole fruit,have usually been given up,resulting in the decrease of economic benefit and serious pollution to environment.However,there is a lot of valuable pectin and flavonoids in peels,and it is very beneficial to extract pectin and flavonoids from peels of Akebia trifoliate koiaz.It can increase both economic benefit of peels and decrease environmental pollution. But these studies have not been reported.In this paper,extracting and separating technology on pectin and flavonoids from Akebia trifoliate koiaz peels was studied.The main results were followed:(1)The optimal parameters of extraction for pectin:extracting temperature(95-100℃); pH1.0-1.2;extracting time for 2.0 h;the ratio of solid to liquid,1:25-1:30.In this case,the rate of extracted and unrefined pectin was up to 11.76%;Active carbon was a good decolouring material for extracting solution:0.7-0.9g active carbons were mixed into 100mL pectin extracting solutions for 35min about 65℃;FeCl3 was an optimal precipitant for extracted pectin.In 100mL pectin extracting solution,high pectin production could be gained by adding 5mL FeCl3(20%,pH 3.5 adjusted with ammonia water),and precipitated over night;The optimal desalting ways were followed:2g pectins were put into 75mL desalting solutions(65%ethanol + 4%hydrochloric + 31%water).After 25min,extracted pectins were carefully washed with 100%ethanol;(2)The optimal parameters of extraction for flavonoids:extracting temperature(80℃); extracting time for 3.0h;the ratio of solid to liquid,1:30.In this case,the rate of extracted and unrefined flavonoids is up to 2.35%;AB-8 micro porous resin was an optimal adsorption isotherm for extracted flavonoids: the static maximum adsorbing capacity was 34.8mg.g-1;the dynamic adsorbing capacity was 21.2mg.g-1at the concentration of extracted flavonoids(0.8mg/mL- 1.5mg/mL);The optimal flavonoids eluting solvent was 50%EtOH-H2O,and the flavonoids eluting ratio was up to 85.9%when the flow velocity was 1-2BV.h-1;Extracted flavonoids was found that they could inhabit to Escherichia coli(DH5α) (Gran negtive)and Staphylococcus aureus Staphylococcus aureus(Gran positive).This study opened up a new and productive way for utilizing Akebia trifoliate koiaz peels,economically,ecologically and socially.
