In this thesis,the category status of 6 strain yeasts,which were isolated from koumiss of 4 different regions,was not determined according to its morphology and biochemistry. Genome DNA was extracted and use a pair of primers,named ITS1 and ITS4 amplify 5.8S rRNA and ITS gene from strains respectively. The amplification was purified and connected into the pMD19-T vector. After clone and identification,determined the sequence. Then compared the sequences we obtained with the sequences in GenBank database in order to identify the strains. And phylogenetic dendrogram were established.Combined with analysis of the phyolgenetic tree and partial sequences of 5.8S rDNA and ITS,Y12 was classified as Saccharomyces unisporus. J14 and S33 was classified as Kluyveromyces marxianus. H19 was classified as Issatchenkia orientalis. S30 and S39 was classified as Candida ethanolica. The result of J14 was as same as that of traditional classification before.
