RNAi(RNA interference) is a process of sequence–specific gene silencing mediated by double-stranded RNA(dsRNA). In order to confer enhanced resistance to Bombyx mori nucleopolyhedrovirus on silkworms using RNAi technology, based on ie-1,lef-1 gene,corresponding dsRNA was designed respectively.Both transgene vectors piggyantiIE-neo with ie-1 dsRNA expression box and piggyantiIE-lef1-neo with ie-1 dsRNA, lef-1 dsRNA expression box were conceived.Stable transformation piggyantiIE-neo BmN cells showed resistance to BmNPV,but it is inferior to piggyantiIE-lef1-neo. During late phase of infection ,Both RNAi effects were concealed for the recovery of viral proliferation. Sequence analysis of the insertion site junctions of stable transformation Bm cells by inverse PCR showed that exotic DNA could be inserted into the cell genome by randomly or typically at TTAA transposition site.We have been doing transgene studies on Bombyx mori primarily using piggyantiIE-lef1-neo and get some eggs and silkworms with expressing GFP.
