Actin-depolymerizing factors Cofilin/destrin (AC proteins), which belong to actin-binding protein, are key regulators of microfilament cystoskeleton. AC proteins can sever microfilament, increase the dissociating rate of monomers, recycling of monomers in actin filaments and depolymerizing of microfilament, and regulate the establishment of microfilament cystoskeleton. Thereby, they would influence some physiologic functions about microfilament cystoskeleton for instance cell proliferation, cell migration, apoptosis and embryonic development and so on .by now, there have no report about AC proteins in mammal reproduction.the aim of this study was to investigate the expression of cofilin1,cofilin2 and destrin which belong to AC proteins in the rat ovary duing folliculogenesis,ovulation,luteinization,formation and regression of corpus luteum. by in situ hybridization and northern blot.Duing follicular development after pmsg treatment, the expression result of cofilin1 mRNA was that: cofilin1 mRNA was seen in undiferentiated theca of follicle, and there were signals of cofilin mRNA in heca interna of antral follicle. During luteinization which was induced by pmsg/hcg, the strong signal was on in theca interna of preovulatory follicle while cofilin1 mRNA reached to peak in follicle wall and granular layer during perovulatory;after ovulation, signal of cofilin mRNA keeped on sturdily during luteinization.During formation and regression of corpus luteum after pmsg/hcg treatment, On day 2 after hCG treatment, there was strong expression of cofilin1 mRNA in corepus luteum, on days 4 and 8 the signal weaked markedly, and the expression of cofilin mRNA increased on day 14 . on days 18 and 22 decreased.The result of northern blot showed that, the level of cofilin1 mRNA on days 2 was higher ,on days 4,8 weaked ,on day 14 increased while on days 18 and 22 weaked. The expression pattern of cofilin1 mRNA in the corpus luteum of pregnancy was similar to that after gonadotropin-treated. Cofilin1 mRNA was detected strongly on day 2. In pregnant corpus luteum, on day 4, 9, 13, the signal was decreased gradually. And the apparente cofilin1 mRNA was seen on day 18, 22. The result of northern blot showed that, the level of cofilin1 mRNA on days 2 was high, on day 4 it decreased while that on days 9, 13 was lower, on days 18, 22 increased.Duing follicular development model, the expression result of destrin mRNA was that: destrin mRNA was seen at a low lever in undiferentiated theca of follicle , there were signals of destrin mRNA in theca interna of antral follicle, and the expression increased gradually following follicular development .During luteinization model,destrin mRNA was seen in theca interna of preovulatory follicle ,but destrin mRNA was strongly observed in follicle wall and granular layer during perovulatory; after ovulation, the signal of destrin mRNA keeped on during luteinization. During formation and regression of corpus luteum model,On day 2 after hCG treatment,there was signals of destrin mRNA in corepus luteum, on days 4 and 8 the signals weaked markedly, and increased on day 14 ,on days 18 ,22 decreased .The result of northern blot showed that: destrin mRNA on days 2 was at a high level, on days 4,8 it weaked, on day 14 increased while on days 18 and 22 decreased. In pregnant corpus luteum, destrin mRNA was detected strongly on day 2, on day 4 decreased, hardly any sigal was seen on day 9and day 13, the apparente destrin mRNA was oberseved on day 18, 22 again. The result of northern blot showed that, the level of destrin mRNA on days 2 was high, on day 4 decreased; and destrin mRNA was hardly detected on days 9, 13 while it increased on days 18, 22.no specific signal of cofilin2 mRNA was detected in the rat ovary duing folliculogenesis,ovulation,luteinization,formation and regression of corpus luteum.In summary, the specific expression of cofilin1 mRNA and destrin mRNA on the undiferentiated theca of follicle and the theca interna suggests that cofilin/destrin may play an important role in differentiation and function of theca folliculi; peaking in expression of cofilin1 mRNA and destrin mRNA on follicle wall and granulose in periovulatory follicles indicates their important role in ovulation and and luteinization. They also were seen duing formation and regression of corpus luteum; it suggests that cofilin1/destrin may take part in formation and regression of corpus luteum.
