Eulaliopsis binata is a valuable apomictic resource in Graminea.In order to study its apomictic characteristic,twelve populations of E.binata were analyzed by Molecular biology,Cytology,Genetics.If the F1 progeny plants could be examined through SSR and RAPD,the molecular markers tightly linked to apomixes and different reproduction plants might be got.Moreover cross-fertilize and Flow Cytomctry can help confirming the mode of reproduction.The results are as follows:1.Establishment of SSR-PCR reaction system in E.binata:2×buffer:2μl;2.0 mmol/L Mg2+;0.2 mmol/L dNTPs;0.3μmol/L SSR primer;80ng DNA和0.5 U Taq enzyme。2.Thirteen primer pairs were developed by Anchor-PCR developing compound SSRs. The development efficiency is about 36%,and six primer pairs of these were polymorphic ones.There polymorphic primer pairs were developed based on searching the genes sequences in public databases.In addition,eighteen SSR primer pairs selected from the relative species were used to analyze.It shows that of 83% primer pairs had product in E.binata,and 56%primer pairs presented polymorphism.3.There are five different progenies were detected by SSR and RAPD:①deficiency bands to parents②supplement bands of parents③toward the maternal bands④toward the paternal bands⑤variation with parents bands.4.The treatments of direct-bagging and emasculation-bagging were as experiment control.Comparing the seed setting of the various treatments,analysis suggested that there were significant differentce among the seed setting of different treatments.So we presumed pollen was necessary to seed to some extent.To E.binata,the frequency of apomixis is very high.5.Using leaves of seeding as control,Xing-zi and Heng-yang populations were screened by Flow Cytometry.At last,two individual plants of Xing-zil4 and Heng-yangl 1 had two peaks 2C:3C,the others had three peaks 2C:3C:4C.So Xing-zil4 and Heng-yang 11 might come from sexual reproduction.
