Culture osteoblast in vitro is an important method in the study of bone metabolism and mechanism of bone formation , it's a tendency that using tissue projecting bone to treat bone defect in the future. The experiment draw the materials from bone and periost in osteoblast culture, these years people pay close attention to using bone possessing potency forming bone to culture osteoblast.Osteoblast is a stub, protuberant cell, it's cytoplasm is basophilous. When the bone is forming , osteoblast spreads to monolayer, connect with each other through apophysis. The main function of osteoblast is generate the fiber of bone and organic matter. After forming intercellular substance of organism, Osteoblast will be burided there ,and become maturate Osteoblast.The experiment chose cranium of new born rat as material, isolating and culture osteoblast use the method of trypsin-collagenase digestion. Obtain the third generation osteoblast, then identify its bionomics. Shape observation, alkaline phosphatase staining,precursor of ground substance staining, nodule of calcification staining, electron microscope observation and so on. After the culture osteoblast form the cranium of new born rat , identify to osteoblast through giemsa staining alkaline,phosphatase staining,alizarin bordeaux staining.
