| To find the influences about Genome and apoptosis on organism after space radiation, 12C6+ particle with different LET(30keV/um and 63.3keV/um) and doses(2mGy, 20mGy,200mGy and 2000mGy) values was adopted to radiate wild Drosophila melanogaster.AFLP with 4 pairs of primers were used in the experiment on the wild Drosophila and the radiated ones.The results showed that:The polymorphic frequency of radiated Drosophila was higher than the wild one;The polymorphic frequency of 63.3keV/um was higher than that of the 30keV/um,the polymorphic frequency enhanced along with the radiation dose in the same LET group;in 2000mGy dose,the difference is the most prominent between different dose groups.In the experiment,13 diverse fragments were isolated and sequenced,6 of them were located on Drosophila chromosomes(5 on the euchromosomes,1 on the X chromosome),3 of fragments belong to transposon,which indicate that transposon may be related with the mutation induced by low dose radiation.In the experiment,RT-PCT was used to analyze the expression of the key apoptosis related genes,including p53,skl,rpr,grim,hid,dIAP1,dIAP2.The results were that: the expression of p53,skl,rpr,grim,hid,in radiated samples were less than the control group;while the expression of dIAP1,dIAP2 is higher than the control.These results showed that the low dose simulation radiation can restrain the apoptosis process of Drosophila. |
