In this paper, we separated, purified and identified the xylon carpohole of Phellinus Nigricans. Based upon colonial morphology on the solid medium and its microscopic mycelian observation , we referred to the《true fungus appreciation dispensatory》and related references to determine its classification: It belongs to Basidiomycetes, Polyporales, Polyporaceace and Phellinus. We also optimized the composition of its culturing medium (including solid and liquid medium) under different conditions. Because it is a kind of wood-rotten mycete, we preliminarily analyzed its exocellular enzyme system, which demonstrated its complete enzyme system to utilize polysaccharides and xylogen. All these provided supportive proof for its future large-scale fermentation.The detail experiment results were listed as follows:1.Through the tissue seperation technique, we seperated and purified the xylon carpohole of Phellinus Nigricans. Brown yellow mycelian colony were obtained from the slant and plate culturing medium. According to microscopic observation about its colonial morphology and mycelia,《true fungus appreciation dispensatory》and related papers helped us to determine its classification: It belongs to Basidiomycetes, Polyporales, Polyporaceace and Phellinus.2.We have optimized the composition of its solid and liquid culturing medium under different conditions. Under the solid condition, we determined the most properiate carbon and nitrogen source are sucrose and wheat bran, which were correspondingly selected from four kinds of carbon source (sucrose, glycose, soluble amylum and lactose) and nine sorts of nitrogen source through one factor and orthogonal design optimization. Meanwhile, the most suitable growth temperatures in solid and liquid medium are 28℃, with most proper pH value as acidic neutral.3.In order to grasp the bioavailability and productivity of the Phellinus Nigricans for the environmental factors in liquid culturing medium, we initially analyzed the the exoenzymatic activities for shelf fungus's four exocellular enzymes. They were cultured for 12 days, which were sampled two days. Single time measurement were carried for all the samples. It was demonstrated that Phellinus Nigricans possess comperatively integral exoenzymes, which allow it to be large-scalely fermented to product the hyphostroma. The most active peak for amylase, laccase and guaiacol enzyme respectively occurred on the sixth, tenth and the fourteenth day, which proved it could degrade the amylum and xylogen. Within the culturing liquid, the crest-time for protein and reducing sugar correspondingly occurred on the tenth and twelfth day that are the proper collection days for protein and revertose.
