Monascus is a kind of filamentous fungus, which can produce different kinds of metabolites such as monascus pigments, Monacolin K, ergosterol, and so on. However, due to the production of citrinin in Monascus, it is called in question about the safety evaluation of Monascus product.In this paper, a genetic engineering strain with the disruption of pksCT gene was constructed by Knock-out, in order to provide consultation for constructing industrialized strain. According to the proteomics method, the proteome of Monascus and its genetic engineering strain were basically researched. The major findings were expounded as follows:1 Disrupted the pksCT gene in Monascus by gene Knock-out, filtrated the converter by using Hph resistance and PCR with gDNA. It had been proved by HPLC that the production of citrinin was reduced in the strain with the disruption of pksCT.2 Extracted total protein in Monascus ruber by TCA-acetone. Established the two-dimensional electrophoresis system by comparing and analyzing the infection caused by different kinds of mediums, component of lysis buffer and the condition of rehydration.3 Analyzed the difference of protein expression between a strain of Monascus ruber with disruption of pksCT and its wild type, the infection to the protein expression had been basically studied.
