In plant development study, it is often necessary to over-express target gene and observe the possible resulted ectopic phenotypes. Besides, it is also necessary to over-express certain gene in complementary verification once the gene has been identified through postional mapping. However, the most generally used consititutive CaMV 35S promoter often fail in some incidences, especially for those genes in male organgenesis and male gametogenesis. Here we linked the promoter sequences of one putative consititutively expression gene ASK1 and CaMV 35S to the cDNA of green fluorescent protein (GFP), respectively. Some transgenic plants had been screened and identified after transforming Arabidopsis plant vis Agobacterium. At the same time, we transformed the two GFP over-expression systems into the Arabidopsis protoplast. We found in 35S::GFP transgenic plant, the expression of GFP driven by CaMV 35S promoter has been found in almost all tissues observed , only with the exception of pollen. Meanwhile we detected no significant difference between the expression of GFP driven by ASK1 promoter and 35S promoter in protoplast. Our work further verified that application of CaMV 35S promoter is limited, especially in plant male orgengenesis and gametogenesis study. So ASK1 can be used as a poromoter in male organgenesis and male gametogenesis.
