Five anaerobic bacteria were isolated from the four samples as follows: sediments of a soda lake in Jilin province,waste water of hot pickled mustard tuber, sediments of Aqike lake in Xinjiang autonomous region and sediments of rich-cobalt region of West Pacific.Based on the 16S rRNA gene analysis,they belong to the four genus of Amphibacillus,Virgibacillus,Aerococcus and Desemzia.Strain Y1T was isolated from sediments of a soda lake in Jilin province and its 16S rRNA gene number in GenBank was FJ169626.Comparative 16S rRNA gene analysis on Eztraon website revealed 93.4-96.8%sequence similarity to members of the genus Amphibacillus.Strain Y1T was facultatively anaerobic,terminal spore-forming and Gram-positive staining rods;not dependent on Na+ but highly halotolerant;the optimum pH for growth was 9.0,with a range of 7.5-10.5;mesophilic with optimum growth at 32℃;be capable of utilizing certain mono- and oligosaccharides and hydrolysing soluble starch and casein;MR-VP tests,catalase and oxidase activities were negative;the predominant fatty acids were branched anteriso-C15:0 and iso-C15:0. It was a member of clusterⅪwithin the low DNA G+C Gram-positive bacterium and the DNA G+C content was 37.7 mol%.Based on the polyphasic analysis,strain Y1T represents a novel species of the genus Amphibacillus,for which the name Amphibacillus jilinensis sp.nov.is proposed.The type strain is Y1T(=CGMCC 1.5123T=JCM 16149T).The other strictly anaerobic and halophilic bacterium isolated from sediments of Xiaokule salt lake in Xinjiang by Kong-Lan Shi,named E2RT was also discussed in this paper.The 16S rRNA gene number of the isolate in GeneBank was FJ169626. Similarity analysis based on the 16S rRNA gene sequences revealed that strain E2RT was most closely related to members of the genus Alkaliphilus with 95.5-91.1% sequence similarity.Further research indicate that strain E2RT represents a novel species of the genus,for which the name Alkaliphilus halophilus sp.nov.is proposed. The type strain is E2RT(=CGMCC 1.5124T=JCM 16124T).
