Since the invention of optical tweezers in 70s the 20th century, peoples attention was focused on it in the field of the study of a variety of micro-particles. Gradient force can be formed by highly focused laser micro-beam. The force can be used to elastically capture particles of biological or other macromolecules, cells, etc in the scope of several nm to tens ofμm. Because of the capability that optical tweezers can manipulate biological samples in vivo without physical access or damage, manipulate optical tweezers technology has become one of the hot spots of the new methods and new instruments of research in current bio-physics. The manipulation using optical tweezers is no mechanical contact of the particle and it possesses suspension features. It can avoid the contact between particles and the sample bottom surface or contact in particles, thus, avoiding the cause of their resistance and interference.In this thesis, the theory of optical tweezers numerical simulation of transverse optical trapping force the design of Single-source optical tweezers experiment system and the measure of cell surface charge experiments are described.In the aspect of quantitative calculations, the axial optical trapping force and lateral optical trapping force on M-particle measurement is calculated quantitatively, the simulation results are described. According to the simulation results, the relations between the optical trapping force and different parameters of the system are discussed. Compared with numerical results of existing theories, we discussed the applicability of this theory on spherical dielectric particles.On the basis of theoretical research results, using optical tweezers instrument, we choose appropriate experiment parameters, measuring the force on yeast cell, and analyze error factor. Then, we measured of cell surface charge of yeast.Finally we generalized and summarized the latest technology in optical tweezers research, a detailed description of some advanced combination of technologies to improve optical tweezers device construction, operation accuracy and calibration in the application was proposed, and on this basis we put forward some ideas and suggestions on it.
