. Microtus fortis is known as the only animal which has the ability of natural resistance to Schistosoma japonicum. It is more used to study mechanisms of resistance to schistosomiasis, and it's little to study as other animal model. We found Microtus fortis is easy prone to fatty liver in the process of experimental animals. To further study the molecular level of pathogenrsis of non-alcoholic fatty liver disease in Microtus fortis. We construct Microtus fortis liver cDNA plasmid library and we get a full-length cDNA sequences of overexpression in Microtus fortis fatty liver, CYP2E1 were obtained.Meanwhile, to verify the role of this gene in the payhogenesis of fatty liver of Microtus fortis. We also constructed a liver-specific PEPCK promoter-driven of PEPCK-CYP2E1-His expression vector. We have established a transgenic technology of testicular injection for Microtus fortis, and we get the transgenic Microtus fortis of integration EGFP gene. Base on this we carry out fatty liver-related gene CYP2E1 transgenic Microtus fortis contruction research, which may lay the foundation for researchies of pathogenesis of non-alcoholic fatty liver disease in Microtus fortis modeles...
