α-amlyase inhibitor is a kind of sugar glucoside hydroltyic enzyme inhibitor.α-amLyase inhibitor separated and purificated from white kidney beans has widely practical value on medicine and agriculture production.This article took white kidney beans as a raw material,α-AI as the object of study, designed experiments to study on the extraction and purification of a-AI from white kidney beans,its properties as well as the comprehensive utilization using leftover got from extraction.The achievements were as follows:1. Had determined the optimum conditions of DNS color method, which used forα-AI experiment analysis method;2. Carries on L9(34) orthogonal test took single factor experiment as foundation, The best extraction process was:white kidney beans grain smashing raw material 100g,screened through 60 mesh sieve, the ratio of material to liquid was 1:6,1.5%NaCl solution extracted over night. centrifugated (10 000rpm,20min 4℃), supernatant 70℃water bath 15min,coldly to the room temperature,added 70% ethyl alcohol 4℃to precipitate. The active average value ofα-AI was 65%(n=3);3.α-AI crude product got from extraction had been eluted by DEAE-Cellulose ionic exchange column chromatographic, the value of a-AI was 0.365g(per 100g raw material), the active value was 79.3%;then Sephadex G-75 gelatin column chromatographic purification got the value ofα-AI was 0.12%, the active value was 90.4%;4. According to PAGE gel electrophoresis(coomassie brilliant blue staining,thymol staining) and Sepharose CL-6B gelatin column chromatographic,α-AI was glycoprotein with structure homogeneous,36kD.The result was basically consistent to the reported in the literatures;5. Lowry method and phenol-sulfuric acid method were used to determined the content of protein and polysaccharide inα-AI sample.Corresponding to the standard curve, the content respectively was 86.7% and 13.3%. 6.α-AI extracted from white kidney beans had high heat proof,a high application value; inhibitory activity was relatively high;7.α-AI extracted from white kidney beans could inhibit a-amylase of mammalian, had inhibitory specificity;8. Explored comprehensive utilization using leftover got from extraction and found the best extraction process for dietary fiber. Concentration of SDF was 37.8%,extraction leftover. quality determination was made for SDF.
