In this study,the fibroblast cells from black bear ear were as the nuclear donors, and the oocytes Yanbian of cattle as the receptor. The different concentrations of taurine, glutamine, glutamic acid, arginine, preserved acid and glycine were added in the basic culture respectively. Then the development of xenogeneic embryos reconstructed was observed, these can determine the most appropriate concentration which plays an active role in the development of the reconstructed heterogeneous embryo in vitro.The results were showed that:In this study, in the basic culture, the test was used three different contnet by 7.0mmol/L,14.0mmol/L,or21.0mmol/L of taurine for the treatment groups and the group without any taurine as the control group to determine the development rate of heterogeneous reconstructed embryo. The results were showed that in the culture culture, the rate of blastocyst development of the taurine concentration for 7.0mmol/L was highest (7.52%), which had a significant difference compared with the other groups, while there is no significant difference on the cleavage rate and developmental rate of 8-16 cells,In this study, in the basic culture, three different concentrations by 0.5mmol/L,1.0mmol/L, or2.0mmol/L of glutamine were treatment group and the control group without glutamine, then the developments rate of heterogeneous reconstructed embryo were determined. The results were showed that the rate of 8-16 cell development L-glutamine concentration for 0.5mmol/L in the culture was 31.26%,that was higher than the other three groups significantly, the highest rate of blastocyst development was 5.27%with a significant difference between the other groups, the cleavage rate had no significant difference.In this study, the basic culture was added 3.2mmol/L,6.4mmol/L,or 12.8mmol/L three different concentrations of glutaminic acid, respectively. Another group without any glutamate was as the control group. The development rates of heterogeneous reconstructed embryo were compared. The results were showed that when glutamate concentration in the culture was 6.4mmol/L, the blastocyst rate was highest for 6.51% with a significant difference compared with the other groups. The cleavage rate and the developmental rate of 8-16 cells, had no significant difference.This study was added with 0.061mmol/L,0.132mmol/L, or 0.264mmol/Lthree different concentrations of arginine in the basic culture and without any arginine as the control group to compare the development rate of heterogeneous reconstructed embryo. The results were showed that in the culture culture the L-arginine concentration of 0.061mmol/L had the highest rate of blastocyst development that was 7.34%with a significant difference between the other groups. There is no significant difference for the cleavage rate and the developmental rate of 8-16 cells,In this study, three different concentrations of proline in the basic culture added 0.143mmol/L,0.285mmol/L, or 0.570mmol/Land the control group was added without proline. The heterogeneous reconstructed embryo development rates were compared. The results were showed that the culture culture with proline concentration (0.143mmol/L) was appeared the highest rate of blastocyst development (5.62%) and had significant differences between other groups. The cleavage rate and developmental rate of 8-16 cells had no significant difference.In this study, in the basic culture,7mmol/L,14mmol/L, or21mmol/L three different concentrations of glycine were added and the control group was without added taurine. The heterogeneous reconstructed embryo development rates were detected, The results were showed that when the taurine concentration of the culture medium wasl4mmol/L, the rate of blastocyst development was highest for 5.73%, significant differences between other groups, The cleavage rate and the developmental rate of 8-16 cells had no significant difference.According to the study on the optimum amino acids concentration comparative, the blastocyst rates with optimal taurine and glutamine concentration group was higher (7.29%,6.87%) than other amino acid groups. The results had significant differences.
