Polysaccharide of Spirulina (Arthrospira) platensis is a kind of natural active substance, it can play an important role in accommodating in the cells. PSP have the marked effect on anti-oxidanting, resisting tumor, resisting radiation and regulating immunity. In the process of making the blood glucose and blood fat lower, PSP also have a good effect.Two kinds of Spirulina ecospecies were treated as materials in this research, one is S.(A.) platensis (Gon) Geitl.(S1) of Erdos, the other is S.(A.) platensis (Gon) Geitl.(S2) in the Lake of Chad. We used water to extract crude polysaccharide from S2 powder, and setup three levels about the extractive time, the extractive temperature, the ratio of solid to water and the pH of the extractive water, in order to obtain optimum parameters condition. The results of the parameters of extracting technology were as follows: the extractive time was 8h,the extractive temperature was 80℃,the ratio of solid to water was 1:25,the pH of extractive water was 12.The results of orthogonal experiment showed that in the process of protein removal, when the concentration of TCA was 8%,the ratio of extractive water of PSP and TCA was 1:1,the number of protein removal were 3,the effectiveness of TCA conditions about protein removal were the best. On the other hand, when the ratio of chloroform and n-butanol was 4:1,the ratio of extractive water of PSP and Sevage solution was 2:1,the number of protein removal were 2, the effectiveness of Sevage conditions were the best to remove protein. The results of experiments were used to weight and scored, we found that the effectiveness of TCA method better than Sevage method in the process of protein removal. And then, we used active carbon to decolorize, and addition quantity of the active carbon, decolorized time, and decolorized temperature have been studied with the orthogonal experiment design method. The results show that the effect of decolorized temperature was the best remarkable element, and the optimized decolorized conditions were decolorized temperature 40℃, the addition quantity of active carbon 1.5% and decolorized time 50 minute.Drosophila melanogaster were used as research animals in this study, it were fed with culture medium which had added PSP in it before.5 experimental groups were desighed to three levels. A was a control group of basic culture medium, B was low-dose group and 0.1% of S2 polysaccharide were added into culture medium, C was medium dose group and 0.3% of S2 polysaccharide were added into culture medium, D was high dose group 0.5% of S2 polysaccharide were added into culture medium, E was medium dose group and 0.3% of S1 polysaccharide were added into culture medium. Then POD,CTA,SOD,GSH-PX and MDA of Drosophila melanogaster were determined to research the antioxidation and mechanism of anti-oxidant of PSP. The results showed that, except the content of MDA had declined, all the other index had heightened, difference between the experiment group and the control group had reached significance level. Therefore, we presume that PSP may improve the activity of a series of antioxidase to reduce the production of the lipid peroxidation, this may be the one path of anti-oxidant and stay caducity.
