Background and purpose"DDK syndrome"shows the DDK males can normally fertilize the females of other strains like in the same strain,while the females of DDK strain are nearly infertile when crossed to males of other strains. Studies supported that the polar infertile characteristic is caused by an incompatibility between the cytoplamic materials in DDK oocyte and sperm facters in other strains,they were all controlled by an alleles of Om locus which has been mapped om mouse Chromosome.Currently in the process of early embryonic development in mice, the main function of the expression of proto-oncogene genes, growth factors, growth and apoptosis genes, such as leukocyte inhibitory factor for several categories, including various functional changes in gene expression would embryos different implications for normal development, and with the Om gene induced embryonic lethal phenomenon in embryology and physiology are very similar, hybrid embryos and the embryo during development of the delivery of candidate gene expression differences can be considered as a direct result of fetal death factor. In this study, using semi-quantitative RT-PCR detected the differences of three lethal hybrid embryos and clarified the contact mechanisms of Om gene and other function genes . Provided a new research ideas for creating a mechanism of egg mutations. Further improved the formation of mouse blastocysts in the theory of multiple gene regulation. The applications of DDK modeland the experimental range are expanded . As a result of non-egg mutation disease of pregnancy to provide a necessary preparation.Materials and Methods In this study, carried out by lethal hybrid embryos (DDK♀x BALB/C♂) as the main study, while the use of normal embryos of the cross (DDK♀x DDK♂) for control of the research work. Used female mice 8-10 weeks of age without producing male and female ratio of 1 : 1 in accordance with mated way male and female mating cage together. Then we selected eight genes EGF, EGF-R, TGF-α, Bcl-2, Mcl-1, C-myc, H-ras, LIF as a candidate gene though microscope observation and collecting eggs at different stages.Using semi-quantitative RT-PCR detected the same gene in different mating combinations in the expression of mouse eggs. Results We collected 90 -110 pieces in morphological development of normal 2 cell to 8 cell stage,and hybrid embryos occurs dispersion phenomenon in 8-cell apoptosis. In the 2 cell stage, EGF, Bcl-2, Mcl-1 are expression differences significant. At 4 cell stage with 6 gene expression, EGF, Bcl-2, Mcl-1, EGF-R 4 genes were differences significant. In 8 cell stage there are seven genes expression, EGF, Bcl-2, Mcl-1, EGF-R 4 genes were significantly different.Conclusion Hybrid embryos from the 8 cell stage occurred after the stage of Mulberry apoptosis, Blastocyst formation due to obstacles, which led to early embryonic death. Candidate genes Mcl-1, Bcl-2, EGF, EGF-R gene in the genetic background of three different periods of embryonic cells expression was significantly different (P <0.01), compared with a normal embryo embryonic hybrid differences in gene expression are lower, the reduction in the expression of these genes may indirectly lead to the phenomenon of early embryonic death.
