A series of porphyrin conjugates have been synthesized,These complexes are studied the interaction with various types of DNA, analyzed the biological activity briefly, and the possible mechanism was elaborated preliminarily.1. Six porphyrin complexes were designed and synthesized.It was confirmed that the products were the target compounds by MS,1HNMR,etc.All the complexes have not been reported in literature.2.The CT-DNA binding properties of these complexes have been investigated by UV- Vis, fluorospectrophotometer,CD spectra and thermal denaturalization. It is found DNA binding properties was influenced by differences of complexes structure. i)the porphyrin linked by ruthenium had a stronger activity.ii)Supported by phen, -based ligand,the binding constant of -OCH3-substituted complexes to CT-DNA is larger than the complexe that of -H-substituted and bpy,dmbpy -based ligand.iii)The order of interaction between complexes based different ligands and CT-DNA is phen > bpy > dmbpy.3. The structural changes of DNA binding properties of these complexes have been investigated by UV-Vis, fluorospectrophotometer and CD spectra. The result showed complex has selective effect on CT-DNA and G-quadruplex.These studies have shown that porphyrin complexes have a rich spectral characteristics, can be selective recognition of biological macromolecules. Generate perturbation of its biological function which indicates its potential applications in pharmaceutical Research.
